Biological Background: CREB3 Function and Localization
- CREB3 (Cyclic AMP-responsive element-binding protein 3), also known as LZIP or LUMAN, is an ER-bound transcription factor belonging to the CREB/ATF family, characterized by an N-terminal transcriptional activation domain and a C-terminal leucine zipper.
- It directly binds to DNA at CRE (5'-GTGACGT[AG][AG]-3') and C/EBP sequences to activate transcription, playing a crucial role in the unfolded protein response (UPR) by promoting cell survival during ER stress. Related references: PMID:10984507, PMID:15845366, PMID:16940180
- Upon ER stress, CREB3 undergoes regulated intramembrane proteolysis (RIP), releasing its N-terminal fragment that translocates to the nucleus to activate UPR target genes such as HERPUD1.
- Subcellularly, CREB3 localizes predominantly to the endoplasmic reticulum membrane and Golgi in resting cells, with nuclear and cytoplasmic accumulation of the cleaved active form after stimulation.
- In addition to UPR, CREB3 positively regulates LKN-1/CCL15-induced leukocyte chemotaxis and is involved in cell proliferation, migration, and differentiation, suggesting tumor-suppressive roles in certain contexts. Related references: PMID:15001559, PMID:17296613, PMID:19779205
- In microbial pathogenesis, CREB3 contributes to HIV-1 protein expression, HCV core-mediated tumor suppression, and HSV-1 latency by sequestering host cell factor HCFC1 at the ER membrane in sensory neurons.
- CREB3 is ubiquitously expressed across tissues, with relatively higher levels in dendritic cells, and its transcriptional activity is fine-tuned by interactions with HDACs and cofactors like CREBZF.
Experimental Guidance and Technical Tips
- This polyclonal antibody was raised against an immunogen spanning amino acids 1–230 of human CREB3; it may detect both full-length and N‑terminal cleavage products.
- For Western blotting (WB), the predicted molecular weight is ~41 kDa, but additional bands corresponding to processed forms may appear; use ER‑stress‑induced cell lysates as positive controls.
- In immunofluorescence (IF/ICC) experiments, permeabilization and induction of ER stress (e.g., with tunicamycin or thapsigargin) can enhance nuclear signal detection.
- For ELISA, validate the antibody with the recombinant immunogen or cell‑based assays, and consider pairing with a detection antibody for sandwich ELISA development.
- The antibody cross-reacts with mouse and rat CREB3, allowing translational studies in rodent models; optimize dilutions and blocking conditions for each species.
- Always include appropriate negative controls (e.g., knockdown or knockout samples) to confirm specificity.
