Biological Background: CRCP Function and Localization
- CRCP (C17/RCP/RPC9) is the DNA-directed RNA polymerase III subunit RPC9, and also serves as an accessory protein for the calcitonin gene-related peptide (CGRP) receptor, where it is known as CGRP-RCP.
- As a core component of RNA polymerase III (Pol III), it participates in the synthesis of small non-coding RNAs including 5S rRNA, tRNAs, snRNAs, and miRNAs from over 500 genomic loci. Related references: PMID:20413673, PMID:33558764, PMID:34675218, PMID:33558766
- Together with POLR3H/RPC8, it forms a mobile stalk that protrudes from the Pol III core and is essential for transcription initiation. Related references: PMID:20413673, PMID:33558764, PMID:33558766
- Pol III acts as a cytosolic and nuclear DNA sensor in innate immunity; it transcribes non-self dsDNA (e.g., viral DNA) into dsRNA, which triggers type I interferon and NF-κB responses via the RIG-I pathway. Related references: PMID:19609254, PMID:19631370
- As CGRP-RCP, it modulates CGRP receptor signaling and cellular responsiveness to CGRP in a variety of tissues.
- The protein localizes to both the nucleus and the cell membrane.
- It is ubiquitously expressed, with the highest levels found in testis.
- Multiple isoforms are generated by alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–100) of human CRCP, which is conserved in the orthologous mouse protein, supporting the observed cross-reactivity.
- For Western blot, the expected molecular weight of human CRCP is approximately 17 kDa (calculated MW ~16.9 kDa). Validate band specificity in your sample type and consider using a positive control (e.g., HeLa or HEK293 cell lysate) alongside a negative control.
- In ELISA, the antibody may be suitable for direct detection of recombinant or native CRCP antigen; optimal coating and detection conditions should be titrated.
- Given its reactivity with mouse, the antibody can be evaluated in mouse model systems for cross-species validation.
