Biological Background: CRBN Function and Localization

  • Protein cereblon (CRBN), encoded by the CRBN gene, is also known as MRT2 or MRT2A. It functions as the substrate recognition component of the DCX (DDB1-CUL4-X-box) E3 ubiquitin ligase complex, targeting specific proteins for ubiquitination and subsequent proteasomal degradation. Related references: PMID:26990986, PMID:33009960
  • Through its ubiquitin ligase activity, CRBN regulates key developmental and signaling processes. It is essential for normal limb outgrowth mediated by fibroblast growth factor FGF8, as shown in studies linking CRBN mutations to congenital limb defects. Related references: PMID:20223979, PMID:24328678, PMID:25043012
  • In the nervous system, CRBN maintains cognitive functions such as memory and learning by negatively regulating large-conductance calcium-activated potassium (BK) channels. It interacts with KCNT1 to control BK channel assembly and neuronal surface expression, influencing presynaptic glutamate release. Related references: PMID:18414909, PMID:29530986
  • CRBN also plays a negative regulatory role in innate immunity by inhibiting Toll-like receptor 4 (TLR4) signaling. It binds TRAF6 and ECSIT, blocking ECSIT ubiquitination, a critical step in TLR4 signal transduction. Related references: PMID:31620128
  • Subcellularly, CRBN is found in the cytoplasm, nucleus, and associated with the membrane. It is widely expressed across tissues but shows highest expression in the brain.
  • Additional features include its annotation as a zinc-binding protein and a phosphoprotein. Alternative splicing produces multiple isoforms, and mutations in CRBN are associated with autosomal recessive intellectual disability.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1-280 of human CRBN (full-length protein ~51 kDa). For Western blotting, consider loading 20-30 µg of protein lysate; the expected band is at approximately 51 kDa under reducing conditions.
  • This antibody is recommended for WB, IHC-P, IF/ICC, and ELISA. For IHC-P, appropriate antigen retrieval methods (e.g., heat-mediated citrate buffer, pH 6.0) may be necessary; validate in tissue sections with known CRBN expression (e.g., brain, limb buds).
  • Cross-reactivity with mouse and rat has been confirmed, suggesting epitope conservation across species. When using these species, optimize conditions and confirm reactivity in your specific experimental model.
  • Given CRBN’s role in the ubiquitin-proteasome system, treatment with proteasome inhibitors (e.g., MG132) may stabilize target proteins and affect CRBN detection in degradation assays.
