Biological Background: CR2/CD21 Function and Localization

  • Complement receptor type 2 (CR2/CD21) serves as a receptor for complement component C3d, facilitating the recognition and uptake of C3d-opsonized antigens by B-cells, thereby enhancing B-cell activation and immune responses. Related references: PMID:1849076, PMID:21527715, PMID:7753047
  • CR2/CD21 forms the B-cell coreceptor complex with CD19, FCRL5, and CD81, which is essential for B-cell signaling and activation. Related references: PMID:1383329, PMID:30107486
  • Independently of the B-cell receptor and CD19, CR2/CD21 can activate SRC kinase, leading to phosphorylation of nucleolin and triggering AKT and GSK3 kinase activities. Related references: PMID:12938232

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 300–590, which includes part of the extracellular domain; consider that the antibody may detect multiple isoforms produced by alternative splicing. Validate in relevant cell lines.
  • For Western blot, expect a major band around 113 kDa; however, due to glycosylation and alternative splicing, additional bands may appear. Optimize sample preparation and gel conditions.
  • For IHC-P and IF-P, target membrane localization requires optimized fixation and permeabilization. Test with known positive tissues such as spleen, tonsil, or B-cell lymphomas.
  • In ELISA, ensure proper coating of recombinant protein or native antigen and include appropriate negative and positive controls.
