Biological Background: CR2/CD21 Function and Localization

  • Complement receptor type 2 (CR2/CD21) is also known as the C3d receptor and Epstein-Barr virus receptor, encoded by the CR2 gene (GeneID: 1380). It belongs to the regulators of complement activation (RCA) family and is involved in innate immunity and host-virus interactions.
  • Functions as a receptor for complement component C3d, HNRNPU, and IFNA1. Binding to C3d-opsonized antigens enhances B-cell antigen uptake and activation, playing a key role in innate immunity (PubMed:21527715, PubMed:1849076, PubMed:7753047). Experimental Guidance and Technical Tips

  • The immunogen corresponds to a synthetic peptide within the C-terminal intracellular domain (amino acids 934–1033). This antibody is expected to detect total CR2/CD21 in Western blot under denaturing and reducing conditions. The predicted molecular weight is ~113 kDa.
  • For immunofluorescence (IF-P), permeabilization may be required to access the intracellular epitope. Optimize fixation and permeabilization conditions for the specific sample type.
  • For ELISA, consider using this antibody as a capture or detection reagent, pairing with another antibody targeting an extracellular epitope if a sandwich format is desired.
  • Validate specificity in the relevant biological context, as cross-reactivity with rat is reported; however, performance in other species should be independently tested.
