Biological Background: CR2/CD21 Function and Localization
- CR2/CD21, also known as Complement receptor type 2, Complement C3d receptor, and Epstein-Barr virus receptor, is a type I membrane glycoprotein.
- It serves as a high-affinity receptor for complement component C3d, attaching to C3d-coated antigens on B-cell surfaces and facilitating antigen recognition and uptake, thereby enhancing B-cell activation and immune responses. Related references: PMID:1849076, PMID:21527715, PMID:7753047
- CR2 forms a B-cell co-receptor complex with CD19, FCRL5, and CD81, which is critical for lowering the threshold of B-cell receptor (BCR) signaling and promoting B-cell activation. Related references: PMID:1383329, PMID:30107486
- Beyond its co-receptor role, CR2 triggers intracellular signaling independently of BCR and CD19 by activating SRC kinase, which phosphorylates nucleolin/NCL, leading to AKT and GSK3 kinase activities. Related references: PMID:12938232
- As a ligand for CD23 (FcεRII), a low-affinity IgE receptor, CR2 participates in regulating IgE production. Related references: PMID:1386409
- CR2 also acts as the receptor for Epstein-Barr virus (EBV) on human B cells, mediating viral entry into host cells.
- The protein is predominantly expressed on the cell membrane of mature B-lymphocytes, and also found on T-lymphocytes, pharyngeal epithelial cells, astrocytes, and follicular dendritic cells of the spleen.
- Post-translational modifications include N-linked glycosylation and disulfide bond formation within the sushi (CCP) domains, characteristic of the complement control protein family.
Experimental Guidance and Technical Tips
- For western blotting, note that the predicted molecular weight of CR2/CD21 is approximately 113 kDa. Consider using a low-percentage SDS-PAGE gel and optimizing transfer conditions for high-molecular-weight proteins.
- In immunohistochemistry (paraffin-embedded tissues), antigen retrieval may be necessary; validate in human lymphoid tissues where CR2 is known to be expressed.
- This recombinant monoclonal antibody offers high specificity and can be employed in ELISA-based assays for detecting human CR2/CD21; consider appropriate blocking and detection protocols.
