Biological Background: CPT2 Function and Localization
- CPT2 (Carnitine O-palmitoyltransferase 2, also known as CPT II) is a mitochondrial enzyme encoded by the CPT2 gene.
- It localizes to the mitochondrial inner membrane, where it plays a pivotal role in fatty acid metabolism.
- CPT2 catalyzes the conversion of acylcarnitines back to acyl-CoA esters, enabling their entry into the beta-oxidation pathway (PubMed:20538056, PubMed:24780397). Related references: PMID:20538056 PMID:24780397
- The enzyme is active with medium-chain (C8-C12) and long-chain (C14-C18) acyl-CoA substrates (PubMed:20538056). Related references: PMID:20538056
- CPT2 is classified as an acyltransferase and belongs to the carnitine acyltransferase family; it is associated with disease variants when deficient.
- Post-translational modifications include acetylation, and a transit peptide directs its mitochondrial import.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 404-658) of human CPT2; this region may be conserved across species, corroborating the reported cross-reactivity with mouse and rat.
- For Western blot, expect a band near 74kDa; employ standard protein extraction and transfer protocols.
- For IHC-P and IF/ICC, consider optimizing antigen retrieval and blocking conditions for your tissue/cell type.
- ELISA may be used for quantitative detection; consider testing the antibody as a capture or detection reagent in a sandwich ELISA setup.
- Since CPT2 is a mitochondrial protein, consider verifying mitochondrial enrichment in your sample preparation.
