Biological Background: CPT1A Function and Localization
- CPT1A (carnitine palmitoyltransferase 1A) is the liver isoform of CPT I, responsible for the transfer of long-chain fatty acyl groups onto carnitine for mitochondrial beta-oxidation. Related references: PMID:11350182, PMID:14517221, PMID:16651524
- Beyond its canonical acyltransferase activity, CPT1A acts as a lysine succinyltransferase, modifying metabolic enzymes such as ENO1 to regulate metabolism independently. Related references: PMID:29425493
- CPT1A supports inducible regulatory T-cell (iTreg) differentiation when activated by butyryl-CoA, which antagonizes malonyl-CoA-mediated repression.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 520-720 of human CPT1A. Use full-length CPT1A lysates or recombinant protein as positive controls for western blot detection.
- For WB, expect a band around 88 kDa (theoretical molecular weight). Validate band identity with appropriate tissue or cell lysates, such as heart or kidney, which express high levels of CPT1A.
- In IHC-P and IF/ICC, mitochondrial outer membrane localization should be observed. Include mitochondrial markers for co-localization studies if needed.
- This antibody cross-reacts with mouse and rat CPT1A based on sequence homology; however, we recommend verifying species-specific reactivity in your experimental system.
- For ELISA, optimize coating antigen and antibody dilution conditions as per your assay platform.
