Biological Background: CPSF73 Function and Localization
- CPSF73 (CPSF3) is a 73 kDa subunit of the cleavage and polyadenylation specificity factor (CPSF) complex, essential for pre-mRNA 3'-end formation via recognition of the AAUAAA signal. Related references: PMID:30507380
- It harbors endonuclease activity, directly cleaving pre-mRNA at the poly(A) site to generate the 3'-hydroxyl group for poly(A) tail addition. Related references: PMID:30507380
- CPSF73 is also critical for histone pre-mRNA 3'-end processing, catalyzing U7 snRNP-dependent endoribonucleolytic cleavage and subsequent 5' to 3' exonuclease degradation of the downstream cleavage product. Related references: PMID:30507380
- Required for progression through the S-phase of the cell cycle; also involved in selective miRNA biogenesis during embryonic stem cell differentiation (By similarity). Related references: PMID:30507380
- Contains zinc-binding motifs essential for its nuclease function and undergoes post-translational modifications including acetylation, ubiquitination, and phosphorylation.
- Localizes predominantly to the nucleus, consistent with its role in RNA processing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within amino acids 585-684 of human CPSF73 (Q9UKF6); epitope conservation among human, mouse, and rat is consistent with the observed cross-reactivity.
- For western blot analysis, expect a band at approximately 77 kDa; optimize loading amounts and lysis conditions for your specific samples.
- This monoclonal antibody is suitable for IP and ELISA; titrate the antibody for each application to achieve optimal signal-to-noise ratio.
- Consider using nuclear extract preparations to enrich target detection, given the nuclear localization of CPSF73.
