Biological Background: CPEB3 Function and Localization

  • CPEB3 is a sequence-specific RNA-binding protein that acts as a translational repressor in the basal unstimulated state but, following neuronal stimulation, acts as a translational activator (By similarity).
  • It binds to a U-rich loop within a stem-loop structure in mRNA 3'-UTRs, distinct from the classical CPE element bound by CPEB1 (By similarity).
  • CPEB3 is essential for the consolidation and maintenance of hippocampal-based long-term memory (By similarity).
  • In basal conditions, CPEB3 represses translation of key synaptic plasticity genes, including GRIA2, DLG4, GRIN1, GRIN2A, GRIN2B, and TRPV1; upon activation, it enhances GRIA1 and GRIA2 translation (By similarity).
  • It negatively regulates target mRNA levels by forming a ternary complex with TOB1 and the deadenylase CNOT7/CAF1, leading to mRNA deadenylation and decay. Related references: PMID:21336257
  • CPEB3 binds to and inhibits the transcriptional activation activity of STAT5B, thereby repressing EGFR transcription, a mechanism linked to memory performance. Related references: PMID:20639532
  • Unlike CPEB1, CPEB2, and CPEB4, CPEB3 is not required for cell cycle progression. Related references: PMID:26398195
  • It localizes to the cytoplasm, nucleus, synapse, dendrites, and postsynaptic density, consistent with its roles in local translation and transcriptional regulation.
  • Keywords include alternative splicing, methylation, phosphoprotein, and Ubl conjugation, indicating post-translational modifications.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a central region of human CPEB3 (aa 150-400), expected to recognize all known isoforms. For WB, a band at ~76 kDa is anticipated; validate with appropriate positive controls such as brain or hippocampal lysates.
  • IHC-P and IF/ICC may require antigen retrieval and permeabilization; colocalization with synaptic markers can be considered.
  • ELISA can be used for detecting native or recombinant CPEB3, noting cross-reactivity with mouse and rat.
