- It is integral to the electron transport chain, facilitating the reduction of molecular oxygen to water and coupling this to proton translocation across the inner mitochondrial membrane to generate the electrochemical gradient for ATP synthesis.
- The COX5B gene encodes a 129-amino acid preproprotein that is processed to a mature ~14 kDa subunit (residues 32–129), which is part of the catalytic core of complex IV.
- Subcellularly, COX5B localizes to the mitochondrial inner membrane.
- The protein contains a zinc-binding site and is subject to post-translational modifications including acetylation, as identified by proteomic studies.
