Biological Background: COX2/PTGS2 Function and Localization

  • COX2/PTGS2 (Prostaglandin G/H synthase 2) is an inducible dual-function enzyme that catalyzes the cyclooxygenation of arachidonic acid to prostaglandin G2 (PGG2) and its subsequent peroxidation to prostaglandin H2 (PGH2), the central precursor of all prostanoids. Related references: PMID:16373578 PMID:22942274 PMID:26859324
  • Beyond arachidonate, COX2 oxygenates other polyunsaturated fatty acids such as dihomo-γ-linolenic acid (DGLA) and eicosapentaenoic acid (EPA) to produce alternative prostanoid series, contributing to diverse lipid mediator profiles. Related references: PMID:11939906 PMID:19540099
  • In addition to classical prostanoid synthesis, COX2 participates in the generation of specialized pro-resolving mediators (SPMs) including D-series resolvins from docosahexaenoic acid (DHA) and E-series resolvins from EPA, playing a key role in the resolution of inflammation. Related references: PMID:12391014 PMID:21206090
  • COX2 is predominantly localized to the membranes of the endoplasmic reticulum, microsomes, and both inner and outer nuclear membranes, positioning it at key intracellular sites for eicosanoid signaling.
  • The enzyme requires heme and iron cofactors for its dual activities and is subject to post-translational modifications including acetylation, S-nitrosylation, and disulfide bond formation, which may regulate its function.
  • COX2 metabolizes 2-arachidonoyl glycerol to prostanoid glyceryl esters, a process linked to pain perception, and can also oxygenate linoleic acid to hydroxyoctadecadienoates (HODEs). Related references: PMID:22942274

Experimental Guidance and Technical Tips

  • The immunogen is designed against the C-terminal region (aa 505-604) of human COX2, an area with high sequence homology across human, mouse, and rat, consistent with the reported cross-reactivity.
  • For Western blot, the predicted molecular weight is ~69 kDa; ensure appropriate gel percentage and transfer conditions. Validate with appropriate positive controls (e.g., COX2-induced cell lysates) and consider the inducible nature of the protein.
  • In IHC-P, antigen retrieval may be necessary; optimize retrieval conditions for your specific tissue and fixation method. Including both positive (e.g., inflamed tissue) and negative (COX2-low) controls is recommended.
  • For ELISA, confirm the peptide epitope recognition in your assay format, and consider using recombinant COX2 protein or peptide as a standard if available.
