Biological Background: COX2/PTGS2 Function and Localization
-
Prostaglandin G/H synthase 2 (COX-2) is a dual-function enzyme with cyclooxygenase and peroxidase activities that catalyze the conversion of arachidonic acid to prostaglandin H2 (PGH2), the precursor of prostaglandins and thromboxanes involved in inflammation (PubMed:16373578, PubMed:22942274, PubMed:26859324). Related references: PMID:16373578, PMID:22942274, PMID:26859324
-
COX-2 also metabolizes other polyunsaturated fatty acids, including dihomo-γ-linolenate and eicosapentaenoate, to generate 1- and 3-series prostaglandins, and contributes to the biosynthesis of specialized pro-resolving mediators (resolvins) from omega-3 fatty acids (PubMed:11939906, PubMed:19540099, PubMed:12391014). Related references: PMID:11939906, PMID:19540099, PMID:12391014
-
Subcellularly, COX-2 localizes primarily to the endoplasmic reticulum, microsomes, and nuclear membranes, positioning it to rapidly respond to stimuli.
-
The enzyme is a glycoprotein with a heme cofactor and disulfide bonds, and its activity is modulated by post-translational modifications including acetylation and S-nitrosylation.
-
PTGS2/COX-2 is a member of the prostaglandin G/H synthase family and is classified under the oxidoreductase, peroxidase, and dioxygenase enzyme classes.
Experimental Guidance and Technical Tips
- For Western blotting, a band around 69 kDa is expected, though post-translational modifications such as glycosylation may cause slight shifts in electrophoretic mobility.
- In IHC-P, consider optimizing antigen retrieval conditions (e.g., heat-induced epitope retrieval in citrate buffer) to achieve specific staining.
- Use appropriate positive and negative controls to validate specificity in your sample system; cross-reactivity with mouse and rat has been reported.
- The immunogen spans amino acids 375–604, a region that may be conserved across species; verify performance if used in non-tested organisms.
