Biological Background: Prostaglandin G/H synthase 1 Function and Localization
- COX1 (Prostaglandin G/H synthase 1), encoded by PTGS1, is the constitutive cyclooxygenase isoform primarily responsible for prostaglandin production in the gastric mucosa and platelets.
- It catalyzes the conversion of arachidonic acid to prostaglandin H2 via sequential cyclooxygenase and peroxidase activities, initiating the biosynthesis of all 2-series prostaglandins and thromboxanes (PubMed:7947975). Related references: PMID:7947975
- The enzyme is an integral membrane protein of the endoplasmic reticulum and microsomal membranes, consistent with its role in eicosanoid biosynthesis.
- COX1 is a heme-containing glycoprotein with an EGF-like domain, disulfide bonds, and alternative splicing variants (UniProt keywords).
- In the stomach, COX1-derived prostaglandin E2 contributes to cytoprotection; in platelets, it generates thromboxane A2 to promote aggregation and vasoconstriction.
