Biological Background: COUP-TFII/NR2F2 Function and Localization
- COUP-TFII (COUP transcription factor 2), also known as ARP-1 or NR2F2, is a member of the nuclear receptor subfamily 2 group F and functions as a ligand-activated transcription factor. It is activated by 9-cis-retinoic acid and all-trans-retinoic acid but not by dexamethasone, cortisol, or progesterone.
- The protein regulates apolipoprotein A-I gene transcription by binding to DNA site A. It may also play a critical role in establishing ovary identity during early gonad development. Related references: PMID:29478779
- COUP-TFII contains zinc-finger DNA-binding domains and belongs to the nuclear hormone receptor family, featuring both DNA-binding and ligand-binding domains.
- The protein localizes primarily to the nucleus, consistent with its role as a transcription regulator.
- It is ubiquitously expressed, with notable presence in the stromal cells of developing fetal ovaries.
- Multiple isoforms exist due to alternative splicing, and the protein undergoes phosphorylation, as identified by proteomic studies.
- Keywords associated with COUP-TFII include activator, DNA-binding, metal-binding, zinc-finger, receptor, transcription, and phosphoprotein, reflecting its functional versatility.
Experimental Guidance and Technical Tips
- The immunogen was designed against the N-terminal region (amino acids 5-100) of human COUP-TFII, which may ensure specificity for endogenous protein detection across multiple species.
- For Western blot (WB), consider using whole-cell lysates with standard protocols and validate with positive and negative controls. A band near 46 kDa is expected.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval and antibody concentration to enhance signal-to-noise ratio.
- For immunoprecipitation (IP), this rabbit monoclonal antibody may provide high affinity; confirm complex formation with appropriate buffers and non-denaturing conditions.
- Given the cross-reactivity with mouse and rat, the antibody can be used for preclinical models; validate species-specific reactivity in the relevant sample system.
