Biological Background: CORIN Function and Localization
- CORIN (atrial natriuretic peptide-converting enzyme) is a serine-type endopeptidase belonging to the transmembrane protease serine family. It processes pro-atrial natriuretic peptide (pro-NPPA) and pro-brain natriuretic peptide (pro-NPPB) into their active hormones, ANP and BNP, which regulate blood pressure, natriuresis, diuresis, and vasodilation. Additionally, CORIN promotes trophoblast invasion and spiral artery remodeling in the pregnant uterus.

PMID:22437503 - Two isoforms exist: isoform 1 (canonical) exhibits full endopeptidase activity, while isoform 2 has significantly weaker catalytic function.
- Subcellularly, CORIN is primarily localized at the cell membrane as a single‑span transmembrane protein. It is also secreted into the bloodstream and can be detected in plasma and serum.
- Tissue expression is highest in the heart, specifically in cardiac myocytes. Significant expression is also observed in the pregnant uterus.
- Structural hallmarks include a signal‑anchor, a transmembrane helix, multiple disulfide bonds, N‑linked glycosylation, and a serine protease domain with autocatalytic activation from a zymogen form.
- Mutations in CORIN are associated with cardiomyopathy, underscoring its cardiovascular importance.
- Beyond cardiac function, CORIN may regulate sodium reabsorption in the kidney (by similarity).
Experimental Guidance and Technical Tips
- For Western blotting, consider using a reducing SDS‑PAGE protocol and a positive control such as human heart tissue or cardiac myocyte lysates. The predicted molecular weight is approximately 116 kDa; pre‑stained markers around this range are helpful.
- Since CORIN is a membrane‑associated protease, protein extraction protocols that enrich for membrane fractions may improve detection sensitivity.
- In ELISA applications, the recombinant immunogen (aa 1‑110) can serve as a coating antigen; validate assay conditions with known positive samples (e.g., recombinant CORIN or natural heart extracts).
- Cross‑reactivity with mouse and rat has been reported; however, confirm species‑specific reactivity in your own experimental setup using appropriate tissue controls.
- For studies involving soluble CORIN (shed form), consider using serum or plasma samples, but note that normal plasma levels may be low.
