Biological Background: COQ3 Function and Localization
- COQ3, also known as 3-demethylubiquinol 3-O-methyltransferase, 3-demethylubiquinone 3-O-methyltransferase, or Polyprenyldihydroxybenzoate methyltransferase, is a mitochondrial O-methyltransferase essential for ubiquinone (Coenzyme Q) biosynthesis.
- It catalyzes two non-consecutive O-methylation steps: the conversion of 3,4-dihydroxy-5-(all-trans-decaprenyl)benzoic acid to 4-hydroxy-3-methoxy-5-(all-trans-decaprenyl)benzoic acid and the methylation of 3-demethylubiquinone to ubiquinone.
Related references: PMID:10777520 PMID:38425362 - The enzyme also mediates the methylation of 3-demethylubiquinol-10 to ubiquinol-10, the reduced form of ubiquinone.
Related references: PMID:10777520 - COQ3 localizes to the mitochondrial inner membrane, consistent with its role in the terminal steps of ubiquinone synthesis.
- According to UniProt, COQ3 is acetylated and contains a transit peptide for mitochondrial import.
- It belongs to the class I-like SAM-binding methyltransferase superfamily and is a member of the UbiG/COQ3 methyltransferase family.
- Keywords include Acetylation, Membrane, Methyltransferase, Mitochondrion, Mitochondrion inner membrane, Proteomics identification, Reference proteome, S-adenosyl-L-methionine, Transferase, Transit peptide, Ubiquinone biosynthesis.
- The human COQ3 protein has a molecular weight of approximately 41 kDa (41054 Da).
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 50-369 of human COQ3, covering the catalytic domain, which may result in broad epitope recognition.
- For Western blot, consider using a mitochondrial-enriched lysate and optimizing primary antibody dilution within the range of 1:500-1:2000 to detect the ~41 kDa band.
- For IF/ICC, anticipate mitochondrial staining; co-localization with a mitochondrial marker is recommended for validation.
- For ELISA, the polyclonal nature may provide high sensitivity; perform a titration to determine the optimal coating antibody and conjugate concentrations.
- Validate antibody performance in your specific sample type and include appropriate positive and negative controls.
