Biological Background: COPS8 Function and Localization
- COPS8, also known as COP9 signalosome complex subunit 8, COP9 homolog (hCOP9), or JAB1-containing signalosome subunit 8, is encoded by the COPS8 gene.
- Core function: Component of the COP9 signalosome (CSN) complex, an essential regulator of the ubiquitin conjugation pathway; mediates deneddylation of cullin subunits of SCF-type E3 ligase complexes, reducing their ubiquitin ligase activity.
- The CSN complex also regulates phosphorylation of p53/TP53, c-jun/JUN, IkappaBalpha/NFKBIA, ITPK1, and IRF8/ICSBP, likely through associated kinases CK2 and PKD; CSN-dependent phosphorylation of TP53 and JUN promotes and protects degradation by the ubiquitin system, respectively.
- Subcellular localization: Cytoplasm and nucleus.
- Keywords: Phosphoprotein, alternative splicing, signalosome; also identified by direct protein sequencing and proteomics.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering amino acids 1-209 of human COPS8, providing broad epitope coverage; consider sequence alignment for cross-reactivity assessments.
- In western blot, expect a band around 23 kDa; validate the observed molecular weight in your specific sample system, as post-translational modifications or isoforms may affect migration.
- Cross-reactivity with mouse and rat COPS8 is indicated; verify sequence homology and expression patterns in these species for your experimental context.
- For IF/ICC, optimize fixation and permeabilization to detect both cytoplasmic and nuclear staining; include appropriate controls.
- ELISA can be used for quantitative analysis; consider testing multiple coating and blocking conditions to maximize signal-to-noise ratio.
