Biological Background: COPG2 Function and Localization

  • COPG2 encodes coatomer subunit gamma-2, also known as gamma-2-COP, a component of the coatomer protein complex. The coatomer is a heptameric complex (alpha-COP, beta-COP, beta'-COP, gamma-COP, delta-COP, epsilon-COP, zeta-COP) that plays a crucial role in vesicle transport between the endoplasmic reticulum (ER) and the Golgi apparatus (By similarity).
  • The coatomer complex binds to dilysine motifs on cargo proteins and is required for the formation of COPI-coated vesicles that mediate retrograde transport from the Golgi to the ER, as well as intra-Golgi trafficking (By similarity).
  • COPG2, along with the other coatomer subunits, is essential for maintaining Golgi structural integrity and influences the processing, activity, and endocytic recycling of LDL receptors (By similarity).
  • The protein localizes to the cytoplasm, cytosol, Golgi apparatus membrane, and COPI-coated vesicle membrane, reflecting its dynamic role in membrane trafficking.
  • COPG2 undergoes alternative splicing, producing multiple isoforms, and is subject to phosphorylation, as indicated by the keywords "Alternative splicing" and "Phosphoprotein" in its UniProt record.
  • Additional keywords associated with COPG2 include ER-Golgi transport, Protein transport, and Transport, highlighting its fundamental role in intracellular protein and lipid trafficking.

Experimental Guidance and Technical Tips

  • This polyclonal antibody was raised against a recombinant protein spanning amino acids 542-871 of human COPG2, covering a substantial portion of the C-terminal domain. This region may contain both conserved and unique epitopes, contributing to cross-reactivity with mouse and rat COPG2.
  • For Western blot (WB), a band at approximately 98 kDa is expected for COPG2; however, due to the polyclonal nature and potential alternative splicing isoforms, additional bands may be observed. Use appropriate positive controls (e.g., human, mouse, or rat cell lysates known to express COPG2) and consider using a blocking agent to reduce non-specific binding.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), COPG2 staining is anticipated to localize to the Golgi apparatus and cytoplasmic vesicles. Fixation and permeabilization conditions should be optimized experimentally. Antigen retrieval may be necessary for paraffin-embedded sections.
  • For ELISA, the antibody can be used for detecting COPG2 in samples, but appropriate standards and validation are required.
  • As with any polyclonal antibody, lot-to-lot variability can occur. It is recommended to validate the antibody in your specific experimental system.
