Biological Background: COPB2 Function and Localization

  • Encodes coatomer subunit beta' (beta'-COP/p102), a core component of the COPI vesicle coat complex that binds to dilysine-based cargo motifs and mediates biosynthetic protein transport from the ER through the Golgi, as well as retrograde Golgi-to-ER retrieval.
  • The coatomer complex is required for Golgi membrane budding, and its function is dependent on the small GTPases of the ARF family; the complex also maintains Golgi structural integrity and modulates processing, activity, and endocytic recycling of LDL receptors.
  • COPB2 localizes to the cytosol, Golgi apparatus membrane, and COPI-coated vesicle membranes, consistent with its dynamic role in vesicular trafficking.
  • The protein contains WD repeat domains and coiled-coil regions, and undergoes post-translational modifications including acetylation and phosphorylation.
  • Acts as a receptor for activated protein kinase C epsilon (RACK), binding to Golgi membranes in a GTP-regulated manner.
  • Associated with human diseases: mutations in COPB2 are linked to primary microcephaly (MCPH19) and osteoporosis (OPDD).

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 657–906, a C-terminal region rich in WD repeats; the resulting rabbit monoclonal antibody is expected to recognize an epitope conserved between human and mouse COPB2.
  • For Western blot, the predicted molecular weight of COPB2 is ~102 kDa; use an appropriate molecular weight marker range, and consider a 6–8% SDS-PAGE gel for optimal resolution.
  • ELISA applications may require optimization of coating antigen and antibody concentration due to the recombinant immunogen design.
  • Validate specificity and lack of cross-reactivity with other coatomer subunits in the relevant species and tissue context.
