Biological Background: Complement Factor H Function and Localization
- Complement factor H (CFH), also known as H factor 1, is a 139 kDa glycoprotein secreted mainly by the liver, and to a lesser extent by monocytes, fibroblasts, endothelial cells, and retinal pigment epithelium.
- As the major soluble inhibitor of the complement alternative pathway, CFH prevents uncontrolled complement activation by binding self cell surfaces, accelerating the decay of fluid-phase and surface-bound C3 convertase (C3bBb). Related references: PMID:19503104 PMID:21317894 PMID:26700768
- CFH serves as a cofactor for complement factor I-mediated proteolytic cleavage of C3b into iC3b, thereby irreversibly inactivating the central amplification loop of the complement cascade. Related references: PMID:18252712 PMID:23332154 PMID:28671664
- Beyond complement regulation, CFH engages integrin receptor CR3 (ITGAM) on neutrophils to promote pathogen adhesion and phagocytosis, linking complement control to innate immune cell responses. Related references: PMID:20008295 PMID:9558116
- During Plasmodium falciparum infection in the mosquito midgut, CFH binds gametocyte surfaces and shields the parasite from alternative complement pathway-mediated killing, highlighting a role in host-pathogen interactions.
- CFH contains multiple short consensus repeat (SCR) sushi domains that mediate its ligand-binding functions, and undergoes post-translational modifications including glycosylation, sulfation, and disulfide bond formation.
- Mutations or polymorphisms in the CFH gene are associated with age-related macular degeneration (AMD), atypical hemolytic uremic syndrome (aHUS), and other complement-related disorders, underscoring its clinical significance.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 100–200 within the SCR domains of CFH; consider that the antibody may recognize both native and denatured forms under reducing and non-reducing conditions, but validation in the target sample system is recommended.
- For Western blotting, a band around 139 kDa is expected under reducing conditions; use appropriate positive controls (e.g., human liver lysate or serum) and molecular weight markers.
- The antibody is validated for ELISA; when designing a sandwich ELISA, test different capture/detection antibody pairs and include relevant matrix controls.
- Cross-reactivity is reported for human; for other species, sequence homology should be checked and experimental verification is advised.
- Complement factor H is abundant in serum; consider sample dilution and blocking strategies to minimize high-background issues in serum-based assays.
