Biological Background: Complement factor D Function and Localization
- Complement factor D (CFD), also designated Adipsin, C3 convertase activator, or Properdin factor D, is a serine protease that serves as the initiating enzyme of the alternative complement pathway, a critical component of innate immunity. - The alternative pathway is initiated by spontaneous hydrolysis of complement C3, and CFD-mediated activation generates C3 and C5 convertases, which amplify the complement cascade and promote opsonization, inflammation, and lysis of pathogens. - In Western blot (WB) analysis, a specific band at approximately 27 kDa is anticipated under reducing conditions. Non-reducing conditions may reveal disulfide-linked multimers or conformational variants; consider testing both conditions.
- For ELISA applications, this polyclonal antibody can be used as a capture or detection reagent for CFD in biological fluids such as serum or plasma. Optimal antibody pairings and concentrations should be determined empirically.
- Due to the secreted nature of CFD, sample preparation may require attention to prevent degradation; adding protease inhibitors and processing samples at low temperatures is advisable.
