Biological Background: Complement C3 Function and Localization

• Complement C3 is a central component of the complement system, circulating as a precursor protein that is proteolytically cleaved into functional fragments (C3a, C3b) upon activation.
• C3b, generated by C3 convertase cleavage, covalently attaches to pathogen surfaces via a reactive thioester bond, acting as an opsonin for phagocytosis and forming C5 convertase to activate C5. • In ELISA, the antibody may be suitable for capturing or detecting native C3 from human, mouse, or rat biological fluids; however, optimal dilution and buffer conditions should be determined empirically.
