Biological Background: Complement C3 Function and Localization
- Complement C3 is a central component of the complement system, circulating as a precursor protein that is proteolytically cleaved into functional fragments (C3a, C3b) upon activation.
- C3b, generated by C3 convertase cleavage, covalently attaches to pathogen surfaces via a reactive thioester bond, acting as an opsonin for phagocytosis and forming C5 convertase to activate C5. - The anaphylatoxin C3a mediates local inflammation by binding C3AR1, triggering G protein-coupled receptor signaling, and stimulating mast cells, basophils, macrophages, and neutrophils to release inflammatory mediators and migrate to sites of inflammation. Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 1364–1663) of human and mouse C3, covering part of the C345C domain; this region is present in both the intact precursor and the C3b/iC3b/C3d fragments.
- For Western blot (WB), consider using reducing conditions to detect the ~186 kDa precursor or the ~110 kDa alpha chain, and validate in relevant samples such as human plasma, serum, or cultured hepatocytes.
- In ELISA, the antibody may be suitable for capturing or detecting native C3 from human, mouse, or rat biological fluids; however, optimal dilution and buffer conditions should be determined empirically.
- Since C3 is abundant and highly conserved, ensure proper controls to confirm specificity, especially when working with closely related alpha-2-macroglobulin family members.
