Biological Background: Complement C3 Function and Localization

  • Complement C3 is a large glycoprotein (∼187 kDa) primarily synthesized in the liver and secreted into plasma, with additional production by adipocytes during both fasting and postprandial periods.
  • As a central hub of the complement cascade, C3 is cleaved by C3 convertase to generate C3b, which covalently attaches to pathogen surfaces via its reactive thioester bond (PubMed:6903192). C3b then acts as an opsonin (PubMed:28264884, PubMed:31507604, PubMed:3653927) and assembles the C5 convertase: with C4b and C2b in the classical/lectin pathways (PubMed:12878586, PubMed:18204047, PubMed:2387864), or with factor B (CFB) in the alternative pathway (PubMed:624565, PubMed:6554279). Related references: PMID:6903192, PMID:28264884, PMID:31507604, PMID:3653927, PMID:12878586, PMID:18204047, PMID:2387864, PMID:624565, PMID:6554279
  • The C3a anaphylatoxin peptide, released upon cleavage, binds to its receptor (C3AR1) and triggers G protein-coupled signaling that activates mast cells and basophils to release histamine, cytokines, and chemokines (PubMed:6968751, PubMed:37169960, PubMed:37852260, PubMed:8702752, PubMed:23383423). It is also a potent chemoattractant for macrophages and neutrophils to sites of inflammation (PubMed:23383423, PubMed:342601, PubMed:5778786). Related references: PMID:6968751, PMID:37169960, PMID:37852260, PMID:8702752, PMID:23383423, PMID:342601, PMID:5778786
  • Beyond immunity, Complement C3 functions as an adipogenic hormone (ASP/C3adesArg), stimulating triglyceride synthesis and glucose transport in adipocytes via the C5a receptor (C5AR2) and activation of PLC, MAPK, and AKT pathways (PubMed:10432298, PubMed:15833747, PubMed:16333141, PubMed:11773063, PubMed:12540846, PubMed:19615750). This activity is non‑inflammatory (PubMed:37852260). Related references: PMID:10432298, PMID:15833747, PMID:16333141, PMID:11773063, PMID:12540846, PMID:19615750, PMID:37852260
  • Subcellularly, native C3 is secreted, while activated C3b fragments can deposit on cell surfaces. The protein undergoes key post‑translational modifications including glycosylation, phosphorylation, and formation of a thioester bond essential for covalent attachment to targets.
  • Genetic variations in C3 are associated with age‑related macular degeneration (ARMD9) and atypical hemolytic uremic syndrome (AHUS5), highlighting its clinical significance.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C‑terminal portion (aa 1364–1663) of human C3, located in the mature C3 alpha chain. For immunofluorescence (IF‑P), consider optimizing fixation and permeabilization protocols for secreted or cell‑surface targets.
  • For ELISA applications, this polyclonal antibody may enable sensitive detection of C3 in plasma, serum, or cell culture supernatants. Pre‑validation of dilution factors and standard curves is recommended.
  • The antibody cross‑reacts with mouse and rat, making it potentially useful for interspecies studies. However, experimental verification of reactivity in the relevant sample system is essential.
