Biological Background: Complement C2 Function and Localization
- C2 is the precursor of the catalytic component of the C3 and C5 convertase complexes, essential in the classical, lectin, and GZMK complement systems (PubMed:12878586, PubMed:17027507, PubMed:18204047). Related references: PMID:12878586 PMID:17027507 PMID:18204047
- Following complement activation, C2 is recruited to pathogen surfaces by C4b to form the C3 convertase, or by C3b and C4b to form the C5 convertase (PubMed:6611150, PubMed:6906228). Related references: PMID:6611150 PMID:6906228
- In the C3 convertase, C2 cleaves C3 into C3a anaphylatoxin and C3b opsonin; in the C5 convertase, it cleaves C5 into C5a and C5b, leading to membrane attack complex formation (PubMed:14561755, PubMed:17027507, PubMed:12878586, PubMed:18204047). Related references: PMID:14561755 PMID:17027507 PMID:12878586 PMID:18204047
- C2 is a secreted protein that also localizes to the cell surface.
- The protein contains multiple structural features and modifications, including disulfide bonds, glycosylation, and a serine protease domain.
- Genetic variants in C2 are associated with age-related macular degeneration and other complement-related disorders.
- Alternative splicing generates multiple isoforms of C2.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 453–752 of human C2; verify coverage of your region of interest if targeting specific isoforms or fragments.
- For Western blot, expect a band at approximately 83 kDa; include appropriate positive and negative controls from human serum or cell lysates.
- For ELISA, optimize coating antigen concentration and antibody dilutions; cross‑validation with a secondary detection system is recommended.
- As a polyclonal antibody, evaluate lot-to-lot consistency and specificity in your assay, potentially using purified C2 protein or complement‑depleted sera.
