Biological Background: COMP Function and Localization

  • Cartilage oligomeric matrix protein (COMP), also known as thrombospondin-5 (TSP5), is a member of the thrombospondin family of extracellular matrix glycoproteins.
  • Plays a critical role in maintaining the structural integrity of cartilage by interacting with collagen, fibronectin, and other ECM components.
  • Mediates chondrocyte interaction with the cartilage extracellular matrix through cell surface integrin receptors.
    Related references:
    PMID:16051604 PMID:16542502
  • Acts as a potent suppressor of apoptosis in chondrocytes by blocking caspase-3 activation and upregulating IAP survival proteins (BIRC2, BIRC3, BIRC5 and XIAP).
    Related references:
    PMID:17993464
  • Essential for preserving the contractile/differentiated phenotype of vascular smooth muscle cells (VSMCs) through interaction with ITGA7.
  • Abundantly expressed in cartilage, bone, tendon, ligament, synovium, and blood vessels; expression increases in late-stage osteoarthritis adjacent to the main defect.
  • Contains EGF-like domains, a coiled-coil region, and multiple calcium-binding repeats; forms pentamers stabilized by disulfide bonds.
  • Alternative splicing produces isoforms; mutations in COMP are linked to pseudoachondroplasia (PSACH) and multiple epiphyseal dysplasia.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a conserved region (aa 50-150) of human COMP, which may account for the observed cross-reactivity with mouse and rat. Confirm performance in your relevant sample system.
  • For Western blot, consider using reduced samples to detect the monomeric ~83 kDa band; native COMP forms disulfide-linked pentamers, so non‑reduced conditions may yield higher molecular weight species.
  • For ELISA, coating conditions and detection antibody pairs should be optimized; consider sandwich ELISA formats to capture and detect COMP in biological fluids.
