Biological Background: COMP (Cartilage Oligomeric Matrix Protein) Function and Localization
- COMP, also known as Thrombospondin-5 (TSP5), is a member of the thrombospondin family of extracellular matrix proteins.
- It plays a critical role in maintaining cartilage structural integrity by interacting with collagens, fibronectin, and cell surface integrin receptors (PubMed:16051604, PubMed:16542502). Related references: PMID:16051604, PMID:16542502
- Acts as a potent suppressor of apoptosis in chondrocytes by blocking caspase-3 activation and inducing IAP survival proteins (BIRC2/3/5, XIAP) (PubMed:17993464). Related references: PMID:17993464
- Essential for maintaining vascular smooth muscle cell (VSMC) contractile phenotype via interaction with ITGA7 (By similarity).
- Localized to the extracellular matrix and secreted into the extracellular space.
- Abundantly expressed in cartilage (especially in osteoarthritis), as well as in bone, tendon, ligament, synovium, and blood vessels.
- Contains EGF-like domains and calcium-binding repeats; undergoes alternative splicing and glycosylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 20-160) of human COMP; the antibody may recognize full-length COMP and certain splice isoforms.
- For Western blot, the predicted molecular weight is approximately 83 kDa (calculated 82.8 kDa). Consider using reducing conditions and appropriate controls.
- In immunofluorescence on paraffin-embedded (IF-P) tissues, antigen retrieval and permeabilization steps may be required for optimal detection.
- Cross-reactivity with mouse and rat has been documented; validation in species-specific sample systems is recommended.
- This polyclonal antibody is suitable for ELISA, but optimal working dilutions should be empirically determined.
