Biological Background: COMMD5 Function and Localization

  • COMMD5 (COMM domain-containing protein 5), also known as HCaRG (hypertension-related calcium-regulated gene protein), is a member of the COMMD protein family.
  • It functions as a scaffold protein in the commander complex that is essential for endosomal recycling of transmembrane cargos; the commander complex includes the CCC subcomplex and the retriever subcomplex. Related references: PMID:37172566, PMID:38459129
  • COMMD5 may modulate the activity of cullin-RING E3 ubiquitin ligase (CRL) complexes. Related references: PMID:21778237
  • It negatively regulates cell proliferation and cell cycle G2/M phase transition, likely by transactivating p21/CDKN1A through a p53/TP53-independent signaling pathway.
  • COMMD5 is involved in kidney proximal tubule morphogenesis.
