Biological Background: COMMD5 Function and Localization
- COMMD5 (COMM domain-containing protein 5), also known as HCaRG (hypertension-related calcium-regulated gene protein), is a member of the COMMD protein family.
- It functions as a scaffold protein in the commander complex that is essential for endosomal recycling of transmembrane cargos; the commander complex includes the CCC subcomplex and the retriever subcomplex. Related references: PMID:37172566, PMID:38459129
- COMMD5 may modulate the activity of cullin-RING E3 ubiquitin ligase (CRL) complexes. Related references: PMID:21778237
- It negatively regulates cell proliferation and cell cycle G2/M phase transition, likely by transactivating p21/CDKN1A through a p53/TP53-independent signaling pathway.
- COMMD5 is involved in kidney proximal tubule morphogenesis.
- It down-regulates activation of NF-kappa-B. Related references: PMID:15799966
- Subcellular localization: Predominantly found in the cytoplasm and nucleus.
- Tissue specificity: Highly expressed in heart, stomach, jejunum, kidney, liver, and adrenal gland, with generally higher expression in adult organs than in fetal tissues.
Experimental Guidance and Technical Tips
- The immunogen covers full-length human COMMD5 (aa 1-224); consider verifying cross-reactivity with other species if needed, as the sequence is highly conserved.
- For Western blotting, an expected band around 25 kDa should be validated in relevant sample lysates; include appropriate controls.
- In immunocytochemistry/immunofluorescence, assess subcellular localization; COMMD5 can appear in both cytoplasm and nucleus depending on cell type and conditions.
- For ELISA, optimize antigen coating and antibody dilution using purified recombinant COMMD5 or peptide standards.
