- Collagen XVII, also known as 180 kDa bullous pemphigoid antigen 2 (BP180) and bullous pemphigoid antigen 2, is encoded by the COL17A1 gene and belongs to the collagen superfamily, featuring a transmembrane domain and multiple collagenous repeats.
- It is a key structural component of hemidesmosomes, playing a critical role in the attachment of basal keratinocytes to the underlying basement membrane, thereby contributing to dermal-epidermal cohesion.
- The protein undergoes proteolytic processing to generate a 120 kDa linear IgA disease antigen (LAD-1), which serves as an anchoring filament component and is targeted by autoantibodies in linear IgA bullous dermatosis.
- COL17A1 is primarily localized to cell junctions, specifically hemidesmosomes, and is also found as a secreted protein in the extracellular matrix and basement membrane.
