Biological Background: Collagen alpha-4(IV) Chain Function and Localization

• Encoded by the COL4A4 gene, collagen alpha-4(IV) chain is a type IV collagen that serves as a major structural component of glomerular basement membranes (GBM), contributing to the 'chicken-wire' meshwork together with laminins, proteoglycans and entactin/nidogen.
• Localized to the extracellular matrix and basement membrane; it is secreted into the extracellular space.
• Exhibits tissue-specific expression: at the protein level, it is found in Bruch's membrane, retinal layers (outer plexiform, inner nuclear, inner plexiform, ganglion cell, inner limiting membrane), and around blood vessels of the retina. Colocalized with alpha-3 type IV collagen in kidney, eye, lens capsule, cochlea, lung, skeletal muscle, aorta, and fetal tissues.
