- Collagen alpha-1(I) chain, encoded by the COL1A1 gene, is a key subunit of type I collagen, the most abundant fibrillar collagen in the human body.
- Type I collagen belongs to group I collagens (fibrillar-forming) and assembles into heterotypic fibrils with collagen alpha-2(I), providing tensile strength to connective tissues.
- The protein is synthesized as a precursor with N- and C-terminal propeptides, which are cleaved during maturation to enable fibril formation.
- It is a secreted extracellular matrix protein, localizing to the extracellular space of tendons, ligaments, and bones, where it supports tissue integrity.
- In bone, collagen fibrils composed of COL1A1 are mineralized with calcium hydroxyapatite, conferring rigidity.
