  • For Western blot, a band around 139 kDa is expected; reducing conditions may help resolve collagen monomers, while non-reducing conditions preserve higher-order oligomers. In tissue lysates, consider collagenase pretreatment to confirm specificity.
  • In immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), collagen is predominantly extracellular; adequate permeabilization (e.g., Triton X-100) and antigen retrieval (for paraffin sections) may be required. Co-staining with ECM markers or detection in decalcified bone sections can provide additional validation.
