- Collagen alpha-1(I) chain (COL1A1) is a major component of type I collagen, the most abundant fibrillar-forming collagen in vertebrates. It belongs to the group I collagen family.
- Type I collagen is a heterotrimer consisting of two alpha-1(I) chains and one alpha-2(I) chain, forming the primary structural element of the extracellular matrix (ECM) in connective tissues.
- The protein is secreted into the extracellular space and deposited in the ECM, where it undergoes extensive post-translational modifications, including hydroxylation, glycosylation, and disulfide bond formation, to assemble into mature fibrils.
- Tissue distribution is predominantly in tendons, ligaments, and bones; in bone, the collagen fibrils are mineralized with calcium hydroxyapatite, providing tensile strength and structural integrity.
- Alternative splicing and proteolytic processing generate multiple isoforms; mutations in COL1A1 are associated with connective tissue disorders such as osteogenesis imperfecta, Ehlers-Danlos syndrome, and certain forms of dwarfism.

Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide mapping to residues 1151–1250 of human COL1A1 (NP_000079.2), a region near the C-terminus. This design may favor detection of the full-length procollagen or mature collagen chains depending on sample processing.
- For Western blot, a band around 139 kDa is expected; reducing conditions may help resolve collagen monomers, while non-reducing conditions preserve higher-order oligomers. In tissue lysates, consider collagenase pretreatment to confirm specificity.
- In immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), collagen is predominantly extracellular; adequate permeabilization (e.g., Triton X-100) and antigen retrieval (for paraffin sections) may be required. Co-staining with ECM markers or detection in decalcified bone sections can provide additional validation.
- For ELISA applications, direct or sandwich formats can be explored. Peptide coating or capture by a second antibody may improve sensitivity when working with complex biological matrices such as serum or tissue extracts.
