Biological Background: Collagen I/COL1A1 Function and Localization

  • Collagen alpha-1(I) chain, also known as alpha-1 type I collagen, is a component of type I collagen, which belongs to the group I collagens (fibrillar forming collagens).
  • Type I collagen is the most abundant collagen and a major structural protein of the extracellular matrix, providing tensile strength to tissues.
  • It is secreted into the extracellular space and extracellular matrix, where it assembles into fibrils.
  • Tissue specificity: it forms the fibrils of tendon, ligaments, and bones. In bones, the fibrils are mineralized with calcium hydroxyapatite, contributing to bone strength.
  • Post-translational modifications include hydroxylation, disulfide bond formation, and phosphorylation (as indicated by keywords). It is also a glycoprotein.
  • Mutations in COL1A1 are associated with various connective tissue disorders, including osteogenesis imperfecta, Ehlers-Danlos syndrome, dwarfism, and osteoporosis (as indicated by UniProt keywords).

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide from the C-terminal region (aa 1151–1250). Ensure that the antibody recognizes the full-length protein in native conditions.
  • For western blot, the predicted molecular weight is approximately 139 kDa. Consider using a 6–8% SDS-PAGE gel for adequate separation of high molecular weight proteins.
  • This antibody cross-reacts with human, mouse, and rat, making it suitable for studies across these species. Verify reactivity in your specific sample system.
  • For IHC-P and IF-P, optimize fixation and antigen retrieval protocols for the tissue type of interest, as collagen-rich tissues may require specific permeabilization steps.
  • For ELISA, validate the antibody pair and assay conditions with appropriate positive and negative controls.
