Biological Background: Collagen I/COL1A1 Function and Localization
- Collagen alpha-1(I) chain (COL1A1) is the primary component of type I collagen, the most abundant fibrillar collagen in vertebrates, essential for structural integrity of connective tissues.
- Type I collagen is a member of group I collagen (fibrillar forming collagen), critical for providing tensile strength.
- The protein is predominantly found in the extracellular matrix of bone, tendons, and ligaments; in bone, fibrils are mineralized with calcium hydroxyapatite.
- COL1A1 is secreted and localized to the extracellular space, where it undergoes extensive post-translational processing, including hydroxylation, glycosylation, and disulfide bond formation.
- Mutations in COL1A1 are linked to connective tissue disorders such as osteogenesis imperfecta and Ehlers-Danlos syndrome, highlighting its clinical significance.
- The mature protein is approximately 138.9 kDa and contains repetitive sequences characteristic of fibrillar collagens.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the C-terminal region (aa 1120–1218) of human COL1A1; consider its potential to recognize both pro- and mature forms, and validate in your sample system.
- For Western blotting, expect a band near 139 kDa for full-length COL1A1; additional bands may represent processed or degradation products.
- In IHC-P and IF/ICC, optimize antigen retrieval and fixation as collagen is highly cross-linked; bone or tendon tissues may require decalcification.
- The antibody cross-reacts with mouse and rat COL1A1, making it suitable for rodent models of skeletal or connective tissue research.
- For ELISA, use appropriate controls to avoid non-specific adsorption often seen with collagen.
