Biological Background: COLEC12 Function and Localization
- COLEC12 (Collectin-12) is also known as CL-P1 (Collectin placenta protein 1), SRCL (Scavenger receptor with C-type lectin), and SCARA4 (Scavenger receptor class A member 4).
- Functions as a pattern-recognition scavenger receptor involved in innate immunity, mediating the binding and phagocytosis of Gram-positive and Gram-negative bacteria as well as yeast.
- Recognizes and internalizes oxidatively modified low-density lipoprotein (oxLDL) in vascular endothelial cells, suggesting a role in atherogenesis.
- Exhibits calcium-dependent carbohydrate-binding activity, with affinity for Gal-type ligands, D-galactose, L-/D-fucose, GalNAc, and T/Tn antigens; specifically internalizes GalNAc in nurse-like cells.
- May contribute to amyloid-beta clearance in the brain, with implications for Alzheimer's disease pathology.
- Localizes to the membrane as a transmembrane receptor.
- Highly expressed in placenta; moderate in heart, skeletal muscle, small intestine, and lung; low in brain, colon, thymus, and kidney. In Alzheimer's disease, upregulated in reactive astrocytes and vascular/perivascular cells.
- Structural features include collagen-like domains, a C-type lectin domain, and a signal-anchor transmembrane region, with a predicted molecular weight of ~81.5 kDa (82 kDa observed due to glycosylation).
Experimental Guidance and Technical Tips
- The polyclonal antibody was raised against amino acids 60–270 of human COLEC12, encompassing the collagen-like domain; this region is relatively conserved, explaining cross-reactivity with mouse.
- For Western blot, expect a band near 82 kDa. Glycosylation may cause a slight shift; validate with appropriate controls (e.g., deglycosylation treatment).
- ELISA can be used to detect COLEC12 in biological samples; optimize coating antigen and antibody concentrations for your specific assay format.
- As a transmembrane protein, use membrane protein extraction protocols to enhance detection sensitivity.
- Placenta and heart lysates serve as strong positive controls due to high endogenous expression, while mouse tissues can be used to confirm cross-reactivity.
