- Collagen alpha-4(IV) chain, encoded by the COL4A4 gene, is a major structural component of type IV collagen, which forms a 'chicken-wire' meshwork in basement membranes together with laminins, proteoglycans and entactin/nidogen.
- Type IV collagen heterotrimers consist of alpha chains; alpha-4(IV) is typically found in combination with alpha-3(IV) and alpha-5(IV) chains, forming essential networks in specialized basement membranes.
- The protein is secreted into the extracellular matrix and is a core constituent of glomerular basement membranes (GBM), playing a critical structural role in renal filtration.
- Tissue distribution is highest in kidney, calvaria, neuroretina and cardiac muscle; lower expression in brain, lung and thymus; not detected in liver, skin, ileum, pancreas or adrenal (at protein level).
- In the eye, COL4A4 is expressed in Bruch's membrane, the inner limiting membrane, retinal layers and around blood vessels, contributing to ocular basement membrane integrity.
- Post-translational modifications include hydroxylation of proline and lysine residues, disulfide bond formation and glycosylation, essential for collagen stability and function.

- For Western blot (WB), a major band of approximately 164 kDa is expected in human kidney or retinal tissue lysates; additional bands may reflect post-translational modifications or degradation products.
- In ELISA, optimize the coating concentration and antibody dilution using a standard indirect protocol with recombinant COL4A4 protein or tissue extracts; include appropriate negative controls.
- Validate cross-reactivity with other collagen family members if required, as polyclonal antibodies may exhibit reactivity to conserved collagenous domains.
