Biological Background: Coagulation Factor X Function and Localization
- Factor X (F10), also known as Stuart factor or Stuart-Prower factor, is a vitamin K-dependent glycoprotein synthesized in the liver and secreted into plasma as a zymogen.
- Upon activation, factor Xa converts prothrombin to thrombin in the presence of factor Va, calcium and phospholipid, a critical step in the coagulation cascade. Related references: PMID:22409427, PMID:39880037
- Factor Xa activates protease-activated receptors (PARs) F2R, F2RL1 and F2RL2, triggering pro-inflammatory signaling. It up-regulates expression of PARs and stimulates production of cytokines such as MCP-1/CCL2, IL6, TNF, IL-1β, IL-8, and IL-18 in endothelial cells and cardiac/atrial tissues. Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 235-488 of human F10, covering part of the catalytic domain. This recombinant protein fragment is expected to generate antibodies recognizing both the zymogen and active forms.
- For Western blotting, a band around 55 kDa is anticipated, but note that glycosylation may cause migration differences. Consider validating in relevant human and mouse plasma or tissue lysates.
- For IF/ICC, appropriate fixation and permeabilization protocols should be optimized for the target cell type. The secreted nature of F10 may require stimulation with coagulation triggers for detection in cell culture.
- In ELISA applications, this polyclonal antibody can serve as a capture or detection reagent; pair with a monoclonal antibody for sandwich ELISA development in hemostasis research.
- Given cross-reactivity with mouse, this antibody may be useful in murine models of thrombosis or inflammation; however, confirm species-specific reactivity in your experimental system.
