  • COASY is a bifunctional enzyme that catalyzes two consecutive steps in the biosynthesis of coenzyme A (CoA): the adenylation of 4'-phosphopantetheine and the phosphorylation of dephospho-CoA to CoA.
  • The enzyme is known as NBP (N-terminal kinase-like) and POV-2, and belongs to the CoA synthase family.
  • Alternative splicing produces multiple isoforms; isoform 2 is predominantly expressed in the brain, while the canonical isoform shows broad tissue distribution.
  • COASY localizes to the cytoplasm, cytosol, and mitochondrial matrix, consistent with its role in CoA metabolism across cell compartments.
  • It is expressed in all examined tissues, with highest levels in kidney and liver and lowest in peripheral blood leukocytes. Isoform 2 is mainly detected in the brain.

  • Cross-reactivity with mouse and rat allows multispecies studies, but independent validation in each species is advised.
