Biological Background: CNTF Function and Localization

- The immunogen is a synthetic peptide corresponding to amino acids 24–74 of human CNTF. For WB, the observed band may appear near the predicted 23 kDa, but consider potential post-translational modifications or cleavage that could alter migration.
- In IF/ICC or IF-P, appropriate permeabilization is recommended since CNTF is primarily cytoplasmic and secreted; validate staining in the relevant tissue/cell type.
- For ELISA, pairing with a capture antibody targeting a different epitope may enhance sensitivity; always include relevant controls.
- Cross-reactivity with mouse and rat CNTF is indicated, but sequence homology should be verified for other species.
