Biological Background: CNDP2 Function and Localization
- CNDP2 (CNDP dipeptidase 2), also known as CN2, CPGL, PEPA, or HEL-S-13, is a member of the M20 metallopeptidase family and functions as a cytosolic non-specific dipeptidase.
- It catalyzes the hydrolysis of amide bonds in dipeptides, with broad substrate specificity including L-cysteinylglycine, a key intermediate in glutathione metabolism. Related references: PMID:12473676 PMID:19346245 PMID:25964343
- Under conditions of amino acid and lactate excess, CNDP2 mediates reverse proteolysis to produce N-lactoyl-amino acids such as N-[(S)-lactoyl]-L-phenylalanine, an exercise-induced metabolite regulating food intake and systemic energy balance. Related references: PMID:25964343
- The enzyme plays a role in cell cycle arrest and apoptosis, and its down-regulation in gastric cancer tissues suggests a tumor-suppressive function. Related references: PMID:17121880 PMID:24395568
- CNDP2 is ubiquitously expressed, with elevated protein levels in kidney and liver, and is also present in peripheral blood leukocytes and fetal tissues. Related references: PMID:12473676
- Subcellularly, CNDP2 localizes to the cytoplasm/cytosol, and it undergoes post-translational modifications including acetylation and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal 210 amino acids of human CNDP2 (NP_060705.2), a domain that is well-conserved across species (human, mouse, rat) and likely to generate antibodies recognizing both native and denatured protein; for Western blot, expect a band near 53 kDa.
- Given CNDP2's ubiquitous expression, perform preliminary validation in lysates from human kidney or liver cell lines, and consider subcellular fractionation to enrich for cytosolic protein if signals are weak.
- For ELISA, use recombinant CNDP2 as a coating antigen and optimize antibody concentrations to minimize non-specific binding.
- As a polyclonal antibody, cross-reactivity with related dipeptidases is possible; confirm specificity by siRNA knockdown or blocking peptide competition in your assay system.
