Biological Background: CNBP Function and Localization
- CNBP (CCHC-type zinc finger nucleic acid binding protein), also known as cellular nucleic acid-binding protein or zinc finger protein 9, is a single-stranded DNA-binding protein that preferentially binds the sterol regulatory element (SRE) sequence 5'-GTGCGGTG-3', mediating transcriptional repression. - It also functions as a transactivator of the Myc promoter, linking it to cell proliferation control (By similarity).
- CNBP binds single-stranded RNA in a sequence-specific manner and specifically targets G-rich elements in target mRNA coding sequences, preventing G-quadruplex structure formation and thereby supporting translation. Related references: PMID:28329689
- The protein contains seven CCHC-type zinc finger domains that confer nucleic acid binding and is subject to post-translational modifications including acetylation, methylation, and phosphorylation, which may regulate its activity.

- The immunogen is a synthetic peptide mapping to residues 70–170 of human CNBP (NP_001120668.1). This region is conserved, and the antibody is reported to cross-react with mouse and rat; however, perform sequence alignment if using in other species.
- For Western blotting, a predominant band at ~19 kDa is expected. Additional bands may represent post-translationally modified forms or isoforms. Consider using appropriate blocking and detection conditions.
- In immunohistochemistry (IHC-P), antigen retrieval may be necessary; optimize according to your tissue and protocol. Include positive and negative controls.
