Biological Background: CMKLR1 Function and Localization

  • Chemerin-like receptor 1 (CMKLR1), also designated ChemR23 or DEZ, is a G protein-coupled receptor that functions as a receptor for the adipokine chemerin/RARRES2 and for resolvin E1.
  • Upon ligand binding, CMKLR1 activates G(i)/G(o) and beta-arrestin pathways, leading to intracellular calcium mobilization, MAPK1/3 (ERK1/2) phosphorylation, and PI3K signaling. These cascades regulate immune cell migration, adipogenesis, and angiogenesis (PubMed:27716822). Related references: PMID:27716822
  • In the immune system, CMKLR1 activation by resolvin E1 down-regulates pro-inflammatory cytokine production in macrophages by inhibiting MAPK1/3 and NF-kappa-B pathways, thereby promoting the resolution of inflammation.
  • Additionally, CMKLR1 can serve as a coreceptor for certain SIV and HIV-1 strains, facilitating viral entry.
  • The receptor is localized at the cell membrane and is prominently expressed in developing bone and cartilage, adult parathyroid glands, cardiovascular tissues, brain, kidney, and various lymphoid and myeloid cells, including monocyte-derived macrophages and immature dendritic cells.
  • CMKLR1 undergoes post-translational modifications such as glycosylation and phosphorylation, and is characterized by disulfide bonds, seven transmembrane helices, and alternative splicing.
  • In metabolic tissues, CMKLR1 is highly expressed in differentiating adipocytes and positively regulates adipocyte metabolism, linking it to energy homeostasis and obesity-related inflammation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a peptide within amino acids 250–350 of human CMKLR1, a region that is conserved in mouse and rat, consistent with the documented cross-reactivity. Verify sequence alignment if additional species are tested.
  • For Western blot, a band around 42 kDa is expected; use appropriate gel percentage and transfer conditions. Optimize antibody dilution for your specific sample type and detection system.
  • When performing ELISA, ensure that the coating antigen presents the relevant epitope; this polyclonal antibody may be used for both direct and sandwich ELISA formats with proper optimization.
  • The antibody is supplied on ice packs; upon receipt, store at the recommended temperature (commonly -20°C) and avoid repeated freeze-thaw cycles to maintain stability.
