Biological Background: Clusterin Function and Localization
-
Acts as an ATP-independent extracellular chaperone that prevents aggregation of non-native and stress-induced blood plasma proteins, and inhibits amyloid fibril formation by proteins such as APP, SNCA, and lysozyme. Related references: PMID:11123922 PMID:19535339 PMID:12176985 PMID:17260971 PMID:12047389 PMID:17407782 PMID:17412999
-
Maintains partially unfolded client proteins in a folding-competent state for subsequent refolding by ATP-dependent chaperones like HSPA8/HSC70; binding to cell surface receptors triggers internalization and lysosomal or proteasomal degradation of the chaperone-client complex. Related references: PMID:11123922 PMID:21505792
-
Inhibits the terminal complement pathway by preventing polymerization of the C9 pore component of the membrane attack complex (MAC), thereby protecting cells from complement-mediated cytolysis. Related references: PMID:2780565 PMID:1903064 PMID:2601725
-
Intracellular forms exhibit dual roles in apoptosis regulation: mitochondrial clusterin suppresses BAX-mediated cytochrome c release and stabilizes mitochondrial membrane integrity via HSPA5, whereas interaction with BCL2L1 can activate BAX to promote cell death; the secreted form does not affect intrinsic apoptosis or TNF-induced NF-kappa-B. Related references: PMID:16113678 PMID:17689225 PMID:22689054 PMID:21567405 PMID:24073260
-
Modulates the ubiquitin-proteasome system by interacting with SCF E3 ligase complexes to promote ubiquitination and degradation of targets such as COMMD1 and IKBKB, thereby regulating NF-kappa-B transcriptional activity. Related references: PMID:20068069 PMID:12882985
-
Localization: secreted into body fluids (blood plasma, CSF, milk, seminal plasma) and found intracellularly in the cytoplasm, nucleus, mitochondria, endoplasmic reticulum, and cytoplasmic vesicles.
-
Ubiquitously expressed with highest levels in brain, testis, ovary, liver, and pancreas; also detected in kidney, heart, spleen, and lung.
Experimental Guidance and Technical Tips
- The immunogen encompasses amino acids 314-449 of the Clusterin alpha chain; confirm reactivity with both the full-length precursor and the processed alpha chain in your assay system.
- In Western blot, the full-length Clusterin precursor is predicted at 52 kDa; under reducing conditions, the cleaved alpha (~40 kDa) and beta (~35 kDa) chains may also be observed. Optimize sample preparation and gel conditions.
- For immunohistochemistry (IHC-P), validate antigen retrieval methods and tissue fixation conditions, and include appropriate positive and negative tissue controls.
- This antibody is compatible with ELISA; consider pairing with a detection antibody targeting a different epitope for sandwich ELISA development.
