Biological Background: Clusterin Function and Localization
- Clusterin (apolipoprotein J, TRPM-2, SGP-2) is a multifunctional glycoprotein encoded by the CLU gene; it exists in secreted and intracellular forms due to alternative splicing and post-translational processing.
- As an extracellular chaperone, clusterin prevents aggregation of non-native and stress-denatured proteins, inhibits amyloid fibril formation, and maintains partially unfolded proteins for refolding by other chaperones in an ATP-independent manner. Related references: PMID:11123922, PMID:19535339, PMID:12176985, PMID:17260971, PMID:12047389, PMID:17407782, PMID:17412999
- Binding of clusterin to cell surface receptors triggers internalization and lysosomal/proteasomal degradation of the chaperone-client complex. Related references: PMID:21505792
- It protects cells from complement-mediated cytolysis by inhibiting the assembly of the membrane attack complex (MAC) through prevention of C9 polymerization. Related references: PMID:2780565, PMID:1903064, PMID:2601725
- Intracellular clusterin interacts with SCF E3 ligases to promote ubiquitination and proteasomal degradation of targets such as COMMD1 and IKBKB, and modulates NF-κB transcriptional activity. Related references: PMID:20068069, PMID:12882985
- A mitochondrial form suppresses BAX-dependent cytochrome c release and stabilizes mitochondrial membrane integrity to inhibit apoptosis. Related references: PMID:16113678, PMID:17689225, PMID:22689054
- Subcellular localization includes secretion into biological fluids, as well as presence in cytoplasm, nucleus, mitochondria, endoplasmic reticulum, and cytoplasmic vesicles.
- Clusterin is ubiquitously expressed with high levels in brain, testis, ovary, liver, and pancreas; detected in blood plasma, cerebrospinal fluid, milk, seminal plasma, and colon mucosa.
Experimental Guidance and Technical Tips
- Due to the polyclonal nature and use of a recombinant immunogen, this antibody is expected to detect both precursor and mature forms of clusterin; include appropriate controls and molecular weight markers.
- For Western blotting, optimization of blocking and antibody dilution is recommended; consider running both reducing and non-reducing SDS-PAGE due to the presence of disulfide bonds in clusterin.
- In immunohistochemistry and immunofluorescence, validate with known positive and negative tissue samples; antigen retrieval may enhance detection in formalin-fixed paraffin-embedded sections.
- As clusterin is a secreted glycoprotein, ensure thorough washing to reduce high background in serum-containing samples.
- The reported cross-reactivity with mouse and rat allows for use across common preclinical models; however, verify epitope conservation for quantitative comparisons.
- Always titrate the antibody in your specific assay system to determine the optimal working dilution.
