Biological Background: Calsyntenin-3 Function and Localization

  • Calsyntenin-3 (CLSTN3), also known as Alcadein-beta (Alc-beta) or CDHR14, is a transmembrane protein encoded by the CLSTN3 gene, belonging to the calsyntenin family.
  • Functions as a postsynaptic adhesion molecule that binds presynaptic neurexins to mediate both excitatory and inhibitory synapse formation. Promotes synapse development by associating with both neurexin-alpha and neurexin-beta at the presynaptic membrane. Related references: PMID:25352602
  • Regulates the balance between excitatory and inhibitory synapses by inhibiting formation of excitatory parallel-fiber synapses and promoting formation of inhibitory synapses in the same neuron.
  • Involved in vitamin C uptake via interaction with the ascorbate transporter SLC23A2/SVCT2. Related references: PMID:34673103
  • Forms a complex with APBA2 and APP, stabilizing APP metabolism and enhancing APBA2-mediated suppression of beta-APP40 secretion, likely by retarding intracellular APP maturation. Related references: PMID:12972431
  • An adipose-specific isoform plays a key role in adaptive thermogenesis by inhibiting CIDEA and CIDEC on lipid droplets, preventing lipid droplet fusion, and promoting sympathetic innervation via S100B secretion.
  • Localizes to the postsynaptic cell membrane, endoplasmic reticulum membrane, Golgi apparatus membrane, dendrites, and lipid droplets.
  • Predominantly expressed in brain (especially GABAergic neurons) and kidney, with low levels in heart, skeletal muscle, liver, placenta, pancreas, and lung. An adipose-specific isoform is restricted to brown adipose tissue. Related references: PMID:12498782 , PMID:12972431
  • Post-translational features include calcium-binding domains, glycosylation, a signal peptide, multiple transmembrane helices, and potential ubiquitination. Contains cadherin-like repeats.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 730–850 of human CLSTN3, a region located in the C-terminal extracellular domain. Consider this when interpreting detection in different isoforms or proteolytic fragments.
  • For Western blot, the predicted molecular weight is 106 kDa; validate antibody specificity in the appropriate sample system (e.g., human or mouse brain lysate, kidney tissue, or brown adipose tissue).
  • Because the antibody is cross-reactive with mouse, it may be suitable for rodent models; confirm performance in your experimental setup.
  • For ELISA, optimize coating antigen and detection conditions; cross-reactivity to mouse CLSTN3 should be verified.
  • Glycosylation or other post-translational modifications may cause band shifts; treatment with deglycosylating enzymes can help confirm specificity.
