Biological Background: CLPX Function and Localization

  • ATP-dependent chaperone and unfoldase; part of the mitochondrial ClpXP protease complex, which recognizes, unfolds, and translocates specific protein substrates to the proteolytic subunit CLPP for degradation. Related references: PMID:11923310, PMID:22710082, PMID:28874591
  • Functions as a chaperone in heme biosynthesis by facilitating the incorporation of pyridoxal phosphate cofactor into 5-aminolevulinate synthase, thereby activating ALA synthesis. Related references: PMID:28874591
  • Involved in mitochondrial DNA (mtDNA) nucleoid distribution by regulating mitochondrial transcription factor A (TFAM) activity. Related references: PMID:22841477
  • Also known as caseinolytic mitochondrial matrix peptidase chaperone subunit X; member of the ClpX chaperone family.
  • Localizes to mitochondrion, mitochondrial matrix, and mitochondrion nucleoid.

  • The immunogen corresponds to the C-terminal region (aa 534–633). Consider that the antibody may detect full-length as well as processed forms.
  • For Western blot, the expected band size is approximately 69 kDa. Validate with appropriate positive controls and consider the endogenous expression levels in your sample.
  • For ELISA, the antibody can be explored as a capture or detection reagent. Titration and pairing studies are recommended.
  • Cross-reactivity with mouse and rat has been demonstrated; verify performance in the relevant species and experimental system.
