Biological Background: CLPTM1 Function and Localization
- CLPTM1, also known as cleft lip and palate transmembrane protein 1, is a 76 kDa multi-pass membrane protein of the endoplasmic reticulum (ER).
- It functions as a putative lipid scramblase, potentially facilitating transbilayer lipid movement in the ER membrane (By similarity).
- CLPTM1 binds and retains GABAA receptors in the ER, thereby modulating inhibitory GABAergic neurotransmission by reducing receptor surface expression; altered signaling is implicated in cleft palate (By similarity).
- The protein is required for efficient glycosylphosphatidylinositol (GPI) inositol deacylation in the ER, a critical step for GPI-anchored protein maturation and trafficking. Related references: PMID:29255114
- CLPTM1 may also contribute to T-cell development (By similarity).
- It is widely expressed across tissues.
