Biological Background: CLPTM1 Function and Localization

  • CLPTM1, also known as cleft lip and palate transmembrane protein 1, is a 76 kDa multi-pass membrane protein of the endoplasmic reticulum (ER).
  • It functions as a putative lipid scramblase, potentially facilitating transbilayer lipid movement in the ER membrane (By similarity).
  • CLPTM1 binds and retains GABAA receptors in the ER, thereby modulating inhibitory GABAergic neurotransmission by reducing receptor surface expression; altered signaling is implicated in cleft palate (By similarity).
  • The protein is required for efficient glycosylphosphatidylinositol (GPI) inositol deacylation in the ER, a critical step for GPI-anchored protein maturation and trafficking. Related references: PMID:29255114
  • CLPTM1 may also contribute to T-cell development (By similarity).
  • It is widely expressed across tissues.
  • Post-translational modifications include glycosylation, phosphorylation, and acetylation.
  • Alternative splicing generates multiple CLPTM1 transcript variants.

Experimental Guidance and Technical Tips

  • For Western blotting, CLPTM1 migrates at ~76 kDa. Use an 8-10% SDS-PAGE gel and standard wet transfer conditions; consider extended transfer times for high molecular weight proteins.
  • The immunogen spans the N-terminal 280 amino acids, covering most of the protein's luminal/extracellular domains; this design may enhance detection of both native and denatured forms in WB and ELISA.
  • In ELISA, the recombinant immunogen fragment can be used as a coating antigen for assay development and antibody titration.
  • As CLPTM1 is an integral membrane protein, include mild detergents (e.g., 0.1% Triton X-100) in lysis buffers to ensure efficient solubilization.
  • The antibody shows cross-reactivity with mouse and rat orthologs; validate in the relevant species and tissue type, as expression may vary.
  • Being a polyclonal antibody, it offers robust signal amplification; however, ensure reagent consistency by establishing acceptance criteria for new lots.
