Biological Background: CLPTM1 Function and Localization
- CLPTM1, also known as cleft lip and palate transmembrane protein 1, is a putative lipid scramblase that is widely expressed in human tissues.
- It plays a role in GABAergic transmission by binding and trapping GABAA receptors in the endoplasmic reticulum (ER), thereby reducing their plasma membrane expression and modulating inhibitory neurotransmission; altered GABAergic signaling can contribute to cleft palate (By similarity).
- CLPTM1 may function as a lipid scramblase, translocating lipids between membrane leaflets (By similarity).
- The protein is required for efficient glycosylphosphatidylinositol (GPI) inositol deacylation in the ER, a crucial step for the maturation of GPI-anchored proteins. Related references: PMID:29255114
- It localizes to the ER membrane, consistent with its roles in protein retention and GPI processing.
- Post-translational modifications include acetylation and phosphorylation, and the protein exists in alternatively spliced isoforms; it is a glycoprotein with a molecular weight of approximately 76 kDa.
- CLPTM1 may also play a role in T-cell development (By similarity).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to the N-terminal region (aa 1-100) of human CLPTM1; therefore, the antibody is expected to recognize the full-length protein and possibly N-terminal fragments or isoforms containing this region. For WB, a band near 76 kDa is anticipated.
- For ELISA, consider using the immunogen peptide as a positive control to validate antibody binding.
- Given the exclusive cross-reactivity with human, this antibody is suitable for studies in human cell lines and tissues. For species other than human, validation is recommended.
- For WB applications, optimize lysate preparation and antibody dilution in your experimental system.
