Biological Background: CLPP Function and Localization

• Protein identity and alternative names: CLPP is the ATP-dependent Clp protease proteolytic subunit, mitochondrial. It is also known as Caseinolytic mitochondrial matrix peptidase proteolytic subunit or Endopeptidase Clp, encoded by the CLPP gene.
• Protease function and substrates: As the catalytic component of the mitochondrial ClpXP complex, CLPP degrades various proteins in an ATP-dependent manner. It exhibits low activity without the CLPX chaperone and cleaves substrates such as casein proteins (CSN1S1, CSN2, CSN3) and synthetic peptides (PubMed:11923310). The complex contributes to general mitochondrial protein quality control (PubMed:15522782), and CLPP specifically cleaves PINK1 within the mitochondrion (PubMed:22354088).
