Biological Background: CLPP Function and Localization
- Protein identity and alternative names: CLPP is the ATP-dependent Clp protease proteolytic subunit, mitochondrial. It is also known as Caseinolytic mitochondrial matrix peptidase proteolytic subunit or Endopeptidase Clp, encoded by the CLPP gene.
- Protease function and substrates: As the catalytic component of the mitochondrial ClpXP complex, CLPP degrades various proteins in an ATP-dependent manner. It exhibits low activity without the CLPX chaperone and cleaves substrates such as casein proteins (CSN1S1, CSN2, CSN3) and synthetic peptides (PubMed:11923310). The complex contributes to general mitochondrial protein quality control (PubMed:15522782), and CLPP specifically cleaves PINK1 within the mitochondrion (PubMed:22354088). - Subcellular localization: CLPP is located in the mitochondrion matrix.
- Tissue expression: Predominantly expressed in skeletal muscle, with intermediate levels in heart, liver, and pancreas, and low levels in brain, placenta, lung, and kidney. Detected at the protein level in liver.

- For Western blotting, prepare whole-cell or mitochondrial-enriched lysates from human, mouse, or rat tissues. Expect a specific band near 30 kDa. Include positive controls from tissues with high CLPP expression, such as skeletal muscle or liver.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider using tissue sections or cell lines with documented CLPP expression. Co-staining with a mitochondrial marker is recommended to confirm subcellular localization.
- For ELISA, the antibody can be used as part of a capture or detection system; proper pairing with a complementary antibody should be experimentally validated.
