Biological Background: CLN5 Function and Localization
- Also known as ceroid-lipofuscinosis neuronal protein 5, CLN5 is a lysosomal membrane protein involved in lipid metabolism and protein depalmitoylation.
- Catalyzes the synthesis of bis(monoacylglycero)phosphate (BMP) via transacylation of lysophosphatidylglycerol, playing a key role in intraluminal vesicle formation and cholesterol homeostasis. Related references: PMID:37708259
- Exhibits BMP synthase activity towards various lysophosphatidylglycerol species with preference for longer chain lengths. Related references: PMID:37708259
- Influences retrograde trafficking of lysosomal sorting receptors SORT1 and IGF2R from endosomes to the trans-Golgi network by controlling retromer recruitment. Related references: PMID:22431521
- Regulates the localization and activation of RAB7A, a GTPase required for retromer complex recruitment to endosomal membranes. Related references: PMID:22431521
- Possesses palmitoyl protein thioesterase (S-depalmitoylase) activity in vitro, likely contributing to protein S-depalmitoylation.
- Subcellular localization is primarily at the lysosome membrane.
- Ubiquitously expressed across tissues.
- Mutations in CLN5 are associated with neuronal ceroid lipofuscinosis (CLN5 disease), a neurodegenerative lysosomal storage disorder characterized by progressive neuronal loss.
Experimental Guidance and Technical Tips
- For Western blotting, the predicted molecular weight of CLN5 is approximately 41 kDa; ensure proper gel percentage and transfer conditions for optimal detection.
- In immunohistochemistry (IHC-P), antigen retrieval methods and antibody dilution may require optimization for different tissue types; include appropriate positive and negative controls.
- As a polyclonal antibody, lot-to-lot consistency should be validated, and we recommend titrating the antibody for your specific assay.
- For ELISA, this antibody can be utilized as a capture or detection reagent; perform checkerboard titrations to establish optimal coating and detection concentrations.
